Your best option is to do it hands on either in a lab or otherwise.

That is how I have been doing it.

Labs are really the only way to know GPO. You can use items like the NSA guides for overviews, but there are so many options that you'll probably not remember them all, if merely read.

Cheers,

First, I echo Prezbedard's wise advice about getting hands-on experience. As for specifics, here are some suggestions to help get you started.

Find some practice questions on group policy, and in the process of answering them, work them out on your equipment. For software deployment, you could try to deploy the Administrative tools (adminpak.msi). Then download the network version of a Windows 2000 or XP Service Pack and push that out via GPO. Run MBSA, see its suggestions on auditing, and implement those via GPO. Watch the results on your clients.

Learning this GPO stuff is mandatory for the tests, and you recognize that. When you correctly use GPOs to solve problems in your own network, then your knowledge and confidence will definitely improve.

Hope this helps.
