On the Cisco 2610 ethernet port to nic card on the server do you use a regular straight thru Cat-5 or do you use a crossover cable....

I have a 2621 router and I use a regular Cat5 cable to connect to the HUB. I would assume that if you want to connect directly to the NIC, then you need a crossover cable.

I have never seen this done, so I am not sure it will work.

To connect it to a normal hub it uses a straight thru cable which suggests it is already internally crossed, therefore for a pc use a x-over

Easiest way to figure out which cable to use:
follow this tip.
Each RJ-45 female jack is either marked with "X" or nothing. when connecting two female jacks using a cat 5 patch cable, one of the two female jack muxt be marked with "X". if neither is marked with "X" than use crossover cable. and again if both female jacks are marked with "X" than use crossover cable.
get it!!
